Let’s take a closer look at these hurdles and how we can overcome them, to ensure cell viability during cryopreservation.
What are the key considerations for cryopreservation?
During cryopreservation, the materials and equipment you use are key to the success of freezing the cells safely. Not only to ensure integrity of the process is maintained but also the cost and time implications.
The standard way to cryopreserve cell culture is using liquid nitrogen, however working at liquid nitrogen temperatures (-196°C) can cause a problem. If your cells are frozen too quickly it can lead to ice crystals forming, causing membrane damage and cell death. The use of cryoprotective agents, such as dimethyl sulfoxide (DMSO) are commonly used to reduce the freezing point and allow for a slower cooling rate.
Finding a cryopreservation container that protects your cells
With these risks in mind, it’s important to find a containment solution that is both chemically compatible and flexible at the temperatures used during cryopreservation.
Single-use bioprocess bags are a containment solution universally used in most bio applications. They provide outstanding biocompatibility, chemical compatibility, barrier and strength properties. With the ability to be customised to fit the specific needs of your process, offering flexibility from beginning to end.
Can we safely adopt this trusted approach to meet the needs of cryopreservation? Are cryopreservation bags the answer?
Vials have long been the containment solution of choice when freezing cells. However specially manufactured cryopreservation bags can provide the viability you are looking for. Bags can not only offer a variety of sizes to allow for the storage of higher numbers of cells than can be achieved using vials. But they also provide increased sterility and integrity in a flexible closed system.
Let’s take a closer look...
Cryopreservation bags provide durability, transparency and high flexibility at a broad temperature range of -200°C to +200°C. This outstanding durability allows for storage in freezers or direct immersion in liquid nitrogen. By providing a closed system, it prevents any contamination to the cell culture that can often be seen during the freezing process.
A closed system means there is a lower chance of any liquid nitrogen making its way into the bag, unlike vials which pose a contamination risk during handling, storage and thawing. Vials also have a greater potential to explode when the liquid nitrogen reverts back to its gas form.
The viability of your cells can be assured and the bag can also be used for testing or sampling at any time through sterile tube connections on the bag itself. Overwraps can also provide an additional layer of protection for your valuable cell culture.
